Introduction: Since there are scant data on the mutagenic and carcinogenic potential of a variety of phthalate esters, the Ames assay was used to investigate the mutagenicity of some of these compounds as an approximation of their carcinogenic potential. This paper presents evidence for the mutagenesis of dimethyl phthalate (DMP) and diethyl phthalate (DEP), along with additional studies on the metabolism of DMP and its binding to macromolecules. DMP is a widely used insect repellent which is available to the general public as an active ingredient in commercial products. In its pure form it is used by agricultural workers in doses up to 100 mglkg applied directly to the skin (I) and by the U.S. military in tropical regions of the world.
